APOE4 (Apolipoprotein E4) is a major genetic variant of the APOE gene, which provides instructions for making a protein that helps carry cholesterol and other fats in the bloodstream. It is the strongest genetic risk factor for late-onset Alzheimer's disease, with individuals carrying one or two copies of the allele having a significantly increased likelihood of developing the condition. Beyond Alzheimer's, the APOE4 allele is also associated with an increased risk of cardiovascular disease and cognitive decline.
